Breakthrough under N,N-DMT
Thresholds, bifurcations and plausible mechanisms
The breakthrough as a temporary substitution of the ordinary world by an autonomous phenomenological regime, not mere intensification. It combines the Bayesian loop (REBUS, explosive prediction error), five progressive thresholds and six phenomenological regimes.

The breakthrough under N,N-DMT refers to the crossing of a threshold beyond which the experience is no longer a mere intensification but a temporary substitution of the ordinary world by an autonomous phenomenological regime. It is read here within a cautious explanatory frame, that of predictive coding and the REBUS model, the anarchic brain (Carhart-Harris and Friston, 2019). That frame describes the collapse of high-level priors under psychedelics, but it was formulated on psilocybin and LSD data: it does not deal with DMT and formalizes no thresholds. The five-threshold mapping presented below is a La Honda proposal.
Neurochemical action
N,N-DMT is an agonist at the 5-HT2A receptor, which carries the bulk of the subjective effects. Other documented targets include 5-HT1A, 5-HT2B, whose affinity is comparable to or greater than that at 5-HT2A, and the SERT and VMAT2 transporters, of which it is a substrate.
The receptor-by-receptor breakdown of effects, often put forward, is not established. 5-HT1A adds no affective tone of its own: it acts as a buffer on overall intensity, since blocking it with pindolol amplifies the experience rather than changing its color. Sigma-1 cannot carry the salience or valence attributed to it: its dissociation constant of 14.75 µM sits two orders of magnitude above human plasma concentrations, which range from 0.17 to 1.08 µM after a high intravenous dose. TAAR1 is activated in rodents, but not at concentrations plausible in humans. The result remains a rapid disintegration of perceptual filters and of the internal model.
Bayesian predictive loop (REBUS)
The central frame remains that of explosive prediction error. Under N,N-DMT, priors and descending predictions collapse abruptly, internal signals (sensory and endogenous) flood the estimate, and the inference (the internal model) loses its coherence. The mechanism rests on the collapse of high-level predictions and the takeover by internal content. This is an explanatory candidate, useful but not ontological proof, and it has not been validated on DMT.
Functional consequences
The reorganization of perception is extreme: appearance of autonomous spaces and worlds; intentional entities or presences; dissolution of the ego and rupture of the subject-object coupling; noetic quality (evidence, felt truth); ineffability, the difficulty of describing the experience in ordinary language.
Mapping the thresholds under N,N-DMT

- Threshold 1, loosening of the DMN: intensified vision, vivid textures, but no rupture of the self.
- Threshold 2, plasticity of the real: perceptual systems fall apart, the real becomes plastic and alien, the ego is dismantled.
- Threshold 3, allusive presences: silhouettes, gazes, communication still allusive. This is the breakthrough threshold.
- Threshold 4, breakthrough with entities: another world, coherent and autonomous, near-certain entities (the ordinary becomes the anomaly).
- Threshold 5, unitive collapse: disappearance of entities and of the observer, dissolution of the self, ontological infinity, partial convergence with 5-MeO-DMT.
Typical phenomenological regimes
Several regimes recur: entity encounters (autonomous presences, communication or interaction, lived as real); autonomous realities (coherent spaces, places of their own, the impression of an inhabited elsewhere); insight and noesis (a sense of evidence, of immediate understanding, felt truth); symbolic death and rebirth (dissolution, tipping, reconstruction of meaning); unity without an observer (in some trajectories, no more world, no more entities, no separate witness).
Neuroscientific data and result
The correlates: disintegration of cortical networks (DMN, ECN), increased cross-network integration; a massive rise in brain entropy (EEG and fMRI); EEG signatures of the breakthrough (Timmermann 2019 to 2023, entity or world at breakthrough). The overall result is among the most radical phenomenological collapses, where the internal model of reality is temporarily suspended in favor of an autonomous regime. These perceived contents (entities, spaces, intelligences) remain an open problem.
Duration, tolerance, and precautions
Smoked or vaped, effects begin within tens of seconds, peak at one to two minutes, and the whole episode lasts ten to twenty minutes; integration takes longer. Tolerance is atypical and dissociated depending on what is measured: across four intravenous doses spaced thirty minutes apart, no tolerance to the subjective effects appears while hormonal responses and heart rate blunt, whereas under continuous infusion a moderate acute tolerance is documented. No experimental data document cross-tolerance with 5-MeO-DMT. Precautions matter (an extremely intense experience, essential set and setting, psychotic vulnerability, integration), without offering medical advice. N,N-DMT is present in Psychotria viridis, in Mimosa tenuiflora (synonym: Mimosa hostilis) and in some Acacia, as well as in endogenous traces, and should not be confused with 5-MeO-DMT.
